在植物中產生二倍體配子的TDM基因中的顯性突變的製作方法

本發明涉及:引起在植物中產生2n配子的TDM基因中的顯性突變;包含所述突變的植物;以及它們在植物育種中的應用。本發明還涉及這樣的植物,在該植物中,TDM基因中的顯性突變與以下基因的失活組合:參與植物中減數分裂重組的基因,和參與減數分裂期間著絲粒的單極取向的基因。這些產生未減數分裂配子的植物也可用於植物育種。



背景技術:

減數分裂是有性繁殖的真核生物(諸如,大多數的開花植物)生命周期中的關鍵步驟。

在正常的減數分裂中,染色體首先複製,產生成對的姐妹染色單體。在該輪複製之後,進行兩輪分裂,稱為減數第一次分裂和減數第二次分裂。在減數第一次分裂期間,同源染色體重組並分離在兩個細胞隔室(compartiment)中,這個兩個細胞隔室各自都包含染色體的一個完整的單倍體內容物。在減數第二次分裂中,由減數第一次分裂產生的兩組染色體進一步分裂,使姐妹染色單體分離開。因此,由該分裂產生的四個孢子是單倍體(n),並且攜帶重組的遺傳信息。

相比之下,在二倍體細胞的有絲分裂期間,染色體進行複製,姐妹染色單體分離產生子代細胞,該子代細胞是二倍體(2n)的並且在遺傳學上與原始細胞相同。

已經示出,由減數分裂期間的異常現象得到的異常配子可用於幾種感興趣植物(包括作物)的遺傳改良(供參考,參見例如,Ramanna&Jacobsen,Euphytica 2003,133,3-18)。具體地,2n配子和未減數分裂配子(apomeiotic gamete)可用於產生多倍體植物,或用於使不同倍性水平的植物,例如四倍體作物與其二倍體野生近緣種雜交,以便在植物育種方法中使用它們的遺傳多樣性。它們也可用於遺傳作圖的方法。未減數分裂配子對於產生無融合生殖植物(即能夠從胚珠的母體組織形成種子的植物)也是感興趣的,從而產生作為母本的遺傳克隆的子代。

2n配子(也稱為二倍體配子)是具有體細胞染色體數,而不是配子體染色體數的配子。引起2n配子形成的異常情況特別包括異常胞質分裂,第一次減數分裂或第二次減數分裂的遺漏(skip),或紡錘體異形(供參考,參見V Veilleux,Plant Breeding Reviews,1985,3,252-288,or Bretagnolle&Thompson,New Phytologist,1995,129,1-22)。這些異常情況導致不同類別的未減數的配子。例如,第一次減數分裂的遺漏產生第一次分裂恢復(FDR)的配子,而第二次減數分裂的缺少產生第二次分裂恢復(SDR)的配子。已經報導了在各種植物物種中能夠產生2n配子的許多突變體。然而,還未對這些植物中參與二倍體配子形成的突變進行表徵。

未減數分裂配子是遺傳學上與原始細胞相同的配子,保留親本的所有遺傳信息。未減數分裂配子的產生是無融合生殖的關鍵因素之一(Bicknell&Koltunow,Plant Cell,2004,16,S228-45)。雖然,超過400種植物是無融合生殖的,但這些植物中包括很少的作物物種。此外,通過雜交來引入這一性狀的嘗試已失敗了(「The Flowering of Apomixis:From Mechanisms to Genetic Engineering」,2001;Editor:Savidan et al.;Publisher:CIMMYT,IRD,European Commission DG VI(FAIR),MEXICO,2001.Spillane et al.,Sexual Plant Reproduction,2001,14,179-187)。

迄今為止,已經證實了只有少數基因與2n或未減數分裂花粉的形成有關。

AtPS1(擬南芥平行紡錘體,Arabidopsis thaliana parallel spindles)的失活產生二倍雄孢子,從而產生具有重組遺傳信息的可育的二倍花粉粒和在子代中產生自發三倍體植物(WO 2010/004431;d』Erfurth et al.,PLoS Genet.,2008,4,e1000274)。

TAM(緩慢異步的減數分裂,也稱為CYCA1;2)或OSD1(第二次分裂的消失)的失活導致在減數第一次分裂之後過早地退出減數分裂,從而產生具有重組遺傳信息的二倍體孢子和SDR配子(d』Erfuth et al.,PLoS Genet.,2010,6,e1000989;d』Erfuth et al.,PLoS Biol.,2009,7,e1000124;WO 2010/079432)。

SPO11-1編碼植物中進行有效減數分裂重組所必需的蛋白,並且它的抑制消除重組和配對(Grelon et al.,Embo J.,2001,20,589-600);而Rec8(At2g47980)編碼著絲粒在減數分裂期間的單極取向所必需的蛋白(Chelysheva et al.,J.Cell.Sci.,2005,118,4621-32),並且它的抑制改變染色單體的分離。Atspo11-1突變體經歷不平衡的第一次分裂,隨後進行第二次分裂,產生不平衡的孢子並且是不育的。Atspo11-1/Atrec8雙突變體經歷有絲分裂樣分裂(而不經歷正常的第一次減數分裂),隨後進行不平衡的第二次分裂,產生不平衡的孢子並且是不育的(Chelysheva et al.,J.Cell.Sci.,2005,118,4621-32)。

在osd1/spo11-1/rec8三重突變體中,spo11-1和rec8突變的存在導致有絲分裂樣的第一次減數分裂,並且osd1突變的存在阻止發生第二次減數分裂。因此減數分裂完全替換成有絲分裂,且不會影響隨後的有性生殖過程。因此,將osd1/spo11-1/rec8突變體命名為MiMe(有絲分裂而不是減數分裂)(d』Erfuth et al.,PLoS Biol.,2009,7,e1000124和WO 2010/079432)。由MiMe突變體獲得的孢子和配子在遺傳上與原始細胞相同。

迄今為止,能夠產生2n配子或未減數分裂配子的植物的工程因此限於有限數目的基因。

因此,為了增加可以經修飾以產生高頻的2n配子或未減數分裂配子的基因的數目,需要參與植物中的這些配子形成的其它基因。

TDM(三次分裂突變,THREE-DIVISION MUTANT)基因,也被命名為TDM1、MS5(蛋白雄性不育,PROTEIN MALE STERILE 5)或POLLENLESS 3編碼屬於植物中保守的小蛋白家族的蛋白。擬南芥(Arabidopsis thaliana)的TDM基因的序列能夠在TAIR資料庫中以登錄號At4g20900來獲得,或在GenBank資料庫中以登錄號NC_003075.7來獲得。它編碼434個胺基酸(aa)的蛋白,其序列在所附的序列表中表示為SEQ ID NO:1。

TDM基因被描述為在減數分裂結束時退出減數第二次分裂所需的基因。TDM突變導致多分體(polyad)的形成,以及由於在正常的減數第一次分裂和減數第二次分裂後進入異常的第三次減數分裂而引起的雄性不育。據表明,tdm突變體攜帶突變,該突變產生編碼在其C末端缺少305或112個胺基酸的截短的TDM蛋白的基因(Bulankova et al.,The Plant Cell,2010,22,3791-3803;Cromer et al.,PLoS Genet.,2012,8,e10028652012;Glover et al.,The Plant Journal,1998,15,345-356;Ross et al.,Chrom.Res.,1997,5,551-559;WO/9730581)。

與此相比,如本文所示,本發明人已經確定了TDM基因中的顯性突變,該顯性突變使得在減數第二次分裂之前提前退出減數分裂,從而產生具有重組遺傳信息的二倍體雄SDR配子和二倍體雌SDR配子以及二倍體孢子。此外,他們已經表明,在spo11-1 rec8雙突變體的TDM基因中引入顯性突變產生了MiMe突變體。

本發明人因此鑑定出了在植物中參與2n配子和未減數分裂配子形成的另一個基因。

與參與二倍體配子產生的其它突變相比(這些突變是隱性的,因此需要初生突變體(突變雜合)的額外自花授粉步驟來獲得突變純合的植物),TDM中的顯性的突變不需要這一步驟。攜帶TDM基因中的顯性突變的初生突變體能夠產生2n配子。



技術實現要素:

因此,本發明提供了一種用於獲得產生第二次分裂恢復(SDR)2n配子的植物的方法,

其中,所述方法包括提供基因內包括顯性突變的植物,所述基因在此指TDM基因,編碼在此被指定為TDM蛋白的蛋白;

其中,當所述植物為芸苔屬Brassica spp時,所述蛋白與SEQ ID NO:1的TDM蛋白的第1至286位胺基酸殘基具有至少75%序列一致性;或者,當所述植物不同於芸苔屬Brassica spp時,所述蛋白與所述殘基具有至少30%序列一致性,並且所述蛋白的前60個胺基酸包括在此被指定為TPP/Q模體的模體X1X2X3,其中,X1為蘇氨酸(T),X2為脯氨酸(P),以及X3為脯氨酸(P)或穀氨醯胺(Q);以及

其中,所述顯性突變使所述植物具有產生SDR 2n配子的能力。

在下面的描述中,使用標準的單字母胺基酸代碼。

TDM基因和蛋白質序列能夠在公共資料庫中獲得,諸如但不限於,Plaza資料庫(http://bioinformatics.psb.ugent.be/plaza/)和phytozome網絡門戶http://www.phytozome.net/(phytosome v9.1)。

在使用具有默認參數的T-Coffee(v6.85)(http://toolkit.tuebingen.mpg.de/t_coffee)進行序列比對之後,對SEQ ID NO:1的第1至286位殘基計算TDM蛋白的蛋白序列一致性;使用Bioedit 7.2.5由該比對獲得百分比一致性(Hall,T.A.,1999.BioEdit:a user-friendly biological sequence alignment editor and analysis program for Windows 95/98/NT.Nucl.Acids.Symp.Ser.41:95-98)。

每個物種具有一個或兩個TDM基因,通常具有一個(圖1和圖2,以及表I)。取決於物種,TDM蛋白由約400個胺基酸至約850個胺基酸組成(圖1)。根據圖1中所示出各種被子植物物種的TDM蛋白的比對表明,TDM蛋白的前半部分是保守的。特別地,所有TDM蛋白的前60個胺基酸都包括保守的TPP/Q模體(圖1)。

在使用具有默認參數的T-Coffee(v6.85)(http://toolkit.tuebingen.mpg.de/t_coffee)進行多重序列比對之後,計算各種被子物種的TDM蛋白與SEQ ID NO:1的TDM蛋白的第1至286位殘基的百分比序列一致性。使用Bioedit 7.2.5由該比對獲得一致性矩陣(Hall,TA,1999.BioEdit:a user-friendly biological sequence alignment editor and analysis program for Windows 95/98/NT.Nucl.Acids.Symp.Ser.41:95-98)。結果示於下表I中。

與SEQ ID NO:1的TDM蛋白的第1至286位胺基酸殘基具有至少30%序列一致性的序列與SEQ ID NO:1的TDM蛋白的第1至286位胺基酸殘基具有至少50%的序列相似性。因此,除了芸苔屬Brassica spp.植物以外的其它植物的TDM蛋白可替代地被限定為與SEQ ID NO:1的TDM蛋白的第1至286位胺基酸殘基具有至少50%的序列相似性,並且包括在該蛋白的前60個胺基酸中的TPP/Q模體。

根據本發明產生的SDR 2n配子可用於2n配子的所有常見應用中,例如用於生產多倍體植物,或允許不同倍性水平的植物之間進行雜交。它們也可用於遺傳作圖的方法,例如WO 2006/094774中所公開的「反向子代作圖」的方法中。

根據用於獲得產生第二次分裂恢復2n配子的植物的方法的優選實施方式,當所述植物不同於芸苔屬Brassica spp時,所述蛋白與SEQ ID NO:1的TDM蛋白具有至少35%的序列一致性,並且逐漸優選地,具有至少40%、45%、50%、55%、60%、65%、70%、75%、80%、85%、90%、95%或98%的序列一致性;或者,當所述植物為芸苔屬Brassica spp時,所述蛋白與SEQ ID NO:1的TDM蛋白具有至少80%的序列一致性,並且逐漸優選地,具有至少85%、90%、95%或98%的序列一致性。根據用於獲得產生第二次分裂恢復2n配子的植物的方法的另一優選實施方式,所述TPP/Q模體位於所述蛋白的SEQ ID NO:1的第16~18位的區域中。

根據本發明,所述顯性突變在TDM基因或TDM轉基因的等位基因內,其中,所述TDM基因或轉基因來自任何植物物種,例如表I中提及的那些物種。因此,能夠產生SDR 2n配子的植物能夠通過TDM基因的定向誘變或隨機誘變來獲得,或者通過遺傳轉化來獲得。

根據本發明的用於獲得產生第二次分裂恢復2n配子的植物的方法的優選實施方式,所述方法包括:

提供在TDM基因的等位基因內具有所述顯性突變的植物,所述植物對於該突變是雜合的或純合的。

TDM基因的誘變可以是定向的或隨機的。在隨機誘變(例如通過EMS誘變)之後,篩選期望基因內的突變體。用於高通量誘變和篩選的方法是本領域可知曉的。作為實例,可以提及TILLING(定向誘導基因組局部突變(Targeting Induced Local Lesions IN Genomes),由McCallum et al.,Plant Physiology,2000,123,439-442所述)。利用標準技術進行定向誘變,該標準技術是本領域所已知的並且利用同源重組,優選與核酸酶例如TALEN或CRISPR組合。

根據本發明的用於獲得產生第二次分裂恢復2n配子的植物的方法的另一優選實施方式,所述植物是轉基因植物,並且所述方法包括:

a)用包含DNA構建體的載體轉化至少一個植物細胞,所述DNA構建體包括具有所述顯性突變的TDM基因;

b)培養所述轉化的植物細胞,以使在其基因組中具有轉基因的植物再生,該轉基因包括所述DNA構建體。

DNA構建體包含TDM基因,該TDM基因可來自與其將引入的植物相同的物種,或來自不同的物種。

在TDM基因內的突變中,可以基於對於該突變是雜合的植物的表型性狀,來確定導致產生SDR 2n配子的能力的那些突變:這些植物可以形成至少5%,優選至少10%,更優選至少20%,還更優選至少50%,並且高達100%的雙孢體,作為減數分裂的產物。

可替代地,可通過它們恢復擬南芥(A.thaliana)spo11/rec8雙突變體的可育力的能力,來確定TDM基因內,導致突變體或轉基因植物具有產生SDR 2n配子的能力的顯性突變,其中可育的spo11/rec8/tdm三重突變體對於TDM突變是雜合的,如本申請的實施例中所證實的,擬南芥(A.thaliana)spo11/rec8雙突變體用於篩選顯性突變,隨後將該顯性突變引入到感興趣的植物中。

根據本發明的用於獲得產生第二次分裂恢復2n配子的植物的方法的另一優選實施方式,所述顯性突變包括保守TPP/Q模體的至少一個殘基的突變或由保守TPP/Q模體的至少一個殘基的突變組成。

上述突變可以是置換、插入或缺失,優選置換或缺失。

在更優選的實施方式中,所述顯性突變包括T殘基的突變和/或它相鄰的P殘基的突變,或由T殘基的突變和/或它相鄰的P殘基的突變組成。TP是潛在的磷酸化位點。本申請的實施例證實了T16或P17殘基的突變能夠顯著地提前退出減數分裂。不希望受理論的束縛,本發明人認為鑑於這些結果,TDM通過磷酸化來調節,以確保減數第一次分裂至減數第二次分裂的過渡。

因此,突變有利地是消除所述模體的T殘基磷酸化的突變,即,破壞TP磷酸化位點的突變。

所述突變有利地是所述T殘基和/或P殘基被不同的殘基置換,例如T被A置換,P被L置換。可替代地,所述突變是缺失T殘基和P殘基,並且最終缺失所述T殘基和/或P殘基兩側的額外殘基,例如缺失1個至10個,優選1個至5個,甚至更優選1個或2個殘基。

如上所限定,本發明的另一方面涉及包括TDM基因的DNA構建體,所述TDM基因具有使突變體/轉基因植物具有產生SDR 2n配子的能力的顯性突變。TDM基因可來自與其將引入的植物相同的物種,或來自不同的物種。DNA構建體包括可表達形式的TDM基因。優選地,TDM基因位於啟動子的轉錄控制下,該啟動子在植物細胞中有功能。啟動子可以是TDM基因啟動子,諸如所述TDM基因的內源啟動子,或在植物中有功能的另一啟動子。

在本領域中,具有大量供選擇的適於在植物中表達異源基因的啟動子。

它們可以例如獲自植物、植物病毒或細菌(諸如農桿菌(Agrobacterium))。它們包括:組成型啟動子,即在大多數組織和細胞中以及在大多數環境條件下具有活性的啟動子;以及,組織特異性啟動子或細胞特異性啟動子,僅僅或主要在特定組織或特定細胞類型中具有活性;和,誘導型啟動子,該誘導型啟動子通過物理刺激或化學刺激,諸如由於線蟲感染而產生的物理刺激或化學刺激而被激活。對啟動子進行選擇以便在性母細胞中具有功能。

常用於植物細胞中的組成型啟動子的非限制性實例是花椰菜花葉病毒(CaMV)35S啟動子、Nos啟動子、1,5-二磷酸核酮糖羧化酶/加氧酶(rubisco)啟動子、木薯葉脈花葉病毒(CsVMV)啟動子。

可用於本發明的器官或組織特異性啟動子特別包括能夠賦予減數分裂相關表達的啟動子,諸如DMC1啟動子(Klimyuk&Jones,Plant J,1997,11,1-14)。

本發明的DNA構建體通常還包括轉錄終止子(例如,35S轉錄終止子、胭脂鹼合酶(Nos)轉錄終止子或TDM基因終止子)。

本發明還包括重組載體,所述重組載體包含本發明的DNA構建體。通常,所述重組載體還包括一個或多個標記基因,允許對轉化的宿主進行選擇。

合適載體的選擇,以及用於將DNA構建體插入到其中的方法是本領域普通技術人員所熟知的。載體的選擇取決於預期的宿主以及所述宿主的預期的轉化方法。在本領域中,對於許多植物物種,雙子葉植物或單子葉植物,有多種用於植物細胞或植物的遺傳轉化方法。作為非限制性實例,可提及的是:病毒介導的轉化,通過顯微注射、電穿孔進行的轉化,微粒介導的轉化,農桿菌(Agrobacterium)介導的轉化等。

本發明還提供了包括本發明的重組DNA構建體的宿主細胞。所述宿主細胞可以是:原核細胞,例如農桿菌(Agrobacterium)細胞;或者真核細胞,例如通過本發明的DNA構建體遺傳轉化的植物細胞。構建體可被瞬時表達;它也可被併入到穩定的染色體外複製子中,或整合到染色體中。

本發明人還發現,通過使TDM突變中的顯性突變與兩個基因的失活相組合,來得到產生未減數分裂配子的MiMe突變體,該兩個基因中的一個基因對於減數分裂重組的起始是必需的並且選自spo11-1、SPO11-2、PRD1、PRD2(AT5G57880)、PRD3/PAIR1和DFO(AT1G07060),另一個基因為REC8。

由MiMe突變體產生的未減數分裂配子可以與SDR 2n配子相同的方式,用於生產多倍體植物或用於不同倍性水平的植物的雜交。它們對於生產無融合生殖植物也是感興趣的,即能夠從胚珠的母體組織形成種子的植物,從而產生為母本的遺傳克隆的子代。

因此,本發明的另一目的是用於獲得產生未減數分裂配子的植物的方法,其中所述方法包括:

a)提供植物,所述植物包括如上定義的在TDM基因中的顯性突變;

b)在所述植物中,抑制參與植物中起始減數分裂重組的第一蛋白,所述蛋白選自以下蛋白:

使用在默認參數下的BLASTP程序,計算spo11-1、spo11-2、prd1、PRD2、DFO、PAIR1或REC8蛋白的在此所提供的蛋白序列一致性和相似性的值。使用評分矩陣BLOSUM62,進行相似性計算。

可通過消除、阻斷或降低其功能,或者有利地通過阻止或下調相應基因的表達,來獲得對上述SPO11-1、SPO11-2、PRD1、PRD2、PAIR1、DFO或Rec8蛋白的抑制。在申請WO2010/00431中詳細公開了SPO11-1、SPO11-2、PRD1、PRD2、PAIR1、DFO或Rec8蛋白以及所述蛋白的抑制。

根據本發明的用於獲得能夠產生未減數分裂配子的植物的方法的優選實施方式,所述方法包括:

a)提供在TDM基因的等位基因內具有顯性突變,從而具有產生SDR 2n配子的能力的植物,該植物對於該突變是雜合的;

f)使步驟e)的植物自花授粉,以獲得對於TDM基因內的突變是純合的,對於選自SPO11-1、SPO11-2、PRD1、PRD2、DFO或PAIR1基因的基因內的突變是純合的,以及對於REC8基因內的突變的是純合的植物。

根據本發明的用於獲得能夠產生未減數分裂配子的植物的方法的另一優選實施方式,所述植物是轉基因植物,並且所述方法包括:

實驗步驟

1.生長條件和基因分型

在21℃下,在16小時至18小時光周期和70%相對溼度下,如先前所述在溫室中(Vignard et al.,PLoS Genet.,2007,3,1894-1906)或者如前所述離體在擬南芥Arabidopsis培養基上(Estelle and Somerville,Mol.Genet.,1987,206,200-206)上培養Arabidopsis擬南芥植物。

如先前所述(d』Erfurth et al.,PLoS Biol.,2009,7,e1000124)對spo11-1-3rec8-2植物進行基因分型。如Cromer et al.,PLoS Genet.,2012,8,e1002865所述對tdm-3植物進行基因分型。

2.EMS誘變和突變鑑定

如先前所述(Crismani et al.,Science,2012,336,1588-1590)進行EMS誘變。通過HigSeqTM2000(Illumina)進行全基因組測序。與擬南芥Arabidopsis thaliana TAIR10(哥倫比亞栽培品種)的對照基因組相比,生成SNP列表。

3.細胞學和倍性分析

使用由d'Erfurth等人(PLoS Genet.,2008,4,e1000274)描述的技術,對雄性減數分裂產物進行觀察、染色體分散和倍性測量。

4.定向誘變構建體和植物轉化

使用TDM U(5'-GACATCGGCACTTGCTTAGAG-3';SEQ ID NO:36)和TDM L(5'-GCGATATAGCTCCCACTGGTT-3';SEQ ID NO:37)通過PCR擴增TDM基因組片段。擴增覆蓋ATG前的986個核苷酸和終止密碼子後的537個核苷酸。通過GatewayTM技術(Invitrogen),將PCR產物克隆到pDONR207TM載體(Invitrogen)中,以產生pENTR-TDM;使用Stratagene QuickChangeTM定點誘變試劑盒,根據製造商的說明對pENTR-TDM進行定向誘變。用於產生TDM的突變形式的誘變引物為SEQ ID NO:38至SEQ ID NO:41:

以及它們各自的反向互補引物。為了產生用於植物轉化的雙元載體,使用Gateway TM系統的二元載體pGWB1進行LR重組反應(Nakagawa et al.,Journal of Bioscience and Bioengineering.2007,104,34-41)。對野生型植物以及分離成spo11-1或rec8或tdm-3突變的植物群,使用農桿菌介導的花浸漬法(Clough,S.J.and Bent A.F.,Plant J.,1998,16,735-743)轉化得到的二元載體pTDM,pTDM-P17L,pT16A和pTDM-Y14A。在含有20mg/L潮黴素的瓊脂板上對轉化的植物進行選擇。

實施例1:TDM中的顯性突變導致提早退出減數分裂

為了鑑定控制減數分裂進程的新基因,基於以下觀點來設計遺傳篩選:導致遺漏第二次減數分裂的突變,諸如osd1和cyca1;2/tam將恢復僅在第二次減數分裂中具有不平衡的染色體分離缺陷的突變體的可育力(d』Erfurth et al.,PLoS Biol.,2009,7,e1000124;d』Erfurth et al.,PLoS Genet.,2010,6,e1000989和WO 2010/079432)。這是spo11rec8雙突變體的情況,其中第一次減數分裂類似於有絲分裂(姐妹染色單體平衡分離至相對極),但是第二次分裂是不平衡的且產生非整倍體的配子,因此具有非常有限的可育力(圖5)(Chelysheva et al.,J.Cell.Sci.,2005,118,4621-32)。在OSD1(d』Erfurth et al.,PLoS Biol.,2009,7,e1000124 2009)或CYCA1;2/TAM(d』Erfurth et al.,PLoS Genet.,2010,6,e1000989)中的導致在減數第二次分裂前退出減數分裂的突變,確實能夠恢復spo11-1 rec8的可育力。因此,基於spo11-1 rec8的可育力的恢復來運行遺傳篩選,旨在鑑定出賦予比osd1或tam還相似缺陷的突變體。儘管它們具有減數分裂分離的缺陷,spo11-1 rec8植物產生了經甲基磺酸乙酯(EMS)誘變的足夠的剩餘種子。假定對EMS突變是雜合的M1植物進行自花授粉,並且在5個批次中收穫以產生M2家族。篩選了與spo11-1 rec8非誘變對照相比,可育力增加的約2000個M2家族(400個批次)。

三個批次分離具有增加可育力的植物。基因分型證實,它們是spo11-1 rec8突變體,這表明了其是真正的抑制因子。通過甲苯胺藍染色對雄性減數分裂產物進行的分析示出,在所有三種情況下,可育植物產生孢子的二分體(如在osd1或cyca1;2/tam中所觀察到的),而不是四分體(如在野生型中所觀察到的),表明了在這些植物中並沒有發生第二次減數分裂(圖3)。候選基因(CYCA1;2/TAM和OSD1)的測序確定了CYCA1;2/TAM在三個家族中的兩個中的隱性突變。所確定的突變是外顯子7(TAIR10chr1:29082522C>T)中的剪接位點,和5'UTR區域中的引入上遊框外起始密碼子(TAIR10chr1:29084174G>A)的突變。互補試驗表明它們是等位基因,證實了CYCA1;2中的突變引起二分體表型和可育力的恢復。第三個家族(spo11rec8(s)-40)在OSD1和CYCA1;2中沒有突變,是本研究的重點。

出乎意料地,染色體分散表明四種植物是四倍體的(圖3)。這表明因果突變(causal mutation)是顯性的,並且導致在M1植物的雄性和雌性器官中均產生二倍體配子。與野生型相比,以~100X覆蓋度對兩種姐妹植物的大部分所進行全基因組測序顯示出1144個SNP的存在。然而,僅15個SNP呈現為純合子。這些少數純合子SNP分散在基因組中,表明它們在誘變前就存在於spo11-1rec8株系中,而不是由於EMS誘導的突變所固定的。幾乎所有檢測到的突變都是雜合子這一事實進一步表明因果突變是顯性的。該突變會在M1植物中顯型表達,在與spo11-1 rec8突變組合下,導致二倍體克隆配子的產生(如在spo11-1rec8osd1三重突變體中觀察到的)(MiMe,d』Erfurth et al.,PLoS Biol.,2009,7,e1000124和WO 2010/079432),因此四倍體M2植物中維持了來自M1植物的EMS誘導的突變的雜合性。

然後,在雜合子SNP中尋找候選的因果突變。在這1129個突變中,預測341個突變將影響編碼序列(無義、錯義或剪接位點)。其中,TDM中導致胺基酸變化的突變(TDM-P17L)由於潛在的因果顯性突變而成為良好候選者。之前已證實,TDM對於減數第二次分裂結束時退出減數分裂是必需的。即使在tdm敲除突變體中觀察到的減數分裂缺陷(額外的一輪分裂)與(spo11rec8(s)-40)缺陷顯著不同,TDM中的顯性突變仍然是(spo11rec8(s)-40)中的因果突變的潛在候選者。

為了驗證該假設,產生含有TDM基因(包括啟動子和終止子)的基因組克隆,並使其突變以重新產生在篩選中所鑑定的突變(TDM-P17L),其中TDM基因能夠補充tdm-3突變體(n=8個轉化體,100%四分體)。當引入spo11-1rec8植物中時,TDM-P17L克隆恢復了初代轉化體的可育力(n=2/3,spo11-1 rec8:0.1個種子/果實(n=197),spo11-1 rec8 TDM-P17L#15:25個種子/果實(n=15),spo11-1 rec8 TDM-P17L#67:48個種子/果實(n=10)),並產生二分體(圖4,表II)。這表明TDM中的突變確實是spo11rec8(s)-40中的因果顯性突變。對spo11-1rec8TDM-P17L轉化體中減數分裂染色體分散的分析顯示出,有絲分裂樣的第一次分裂,其中10個單價體在中期I時對齊,並且姊妹染色單體在後期I分離,且不進行第二次分裂(圖5)。接下來,研究了spo11-1rec8 TDM-P17L後代的倍性水平。在自交子代中,僅發現四倍體(4n)(表III)。當spo11-1rec8 TDM-P17L花粉用於使野生型植物受精時,所有得到的子代都是三倍體(表III)。當spo11-1 rec8 TDM-P17L胚珠經野生型花粉粒受精時,僅發現三倍體植物(表III)。這證實了被TDM-P17L轉化的spo11-1 rec8突變體產生高水平的雄性和雌性(100%)有絲分裂樣來源的孢子,其導致功能性二倍體配子。

當引入到野生型植物和tdm-3突變體中時,TDM-P17L基因組克隆通過產生二分體來修飾兩種基因型的減數分裂表型(圖4,表II)。

總之,tdm-p17L顯性突變賦予了與隱性osd1或tam突變類似的減數分裂缺陷,使在第二次分裂之前提前退出減數分裂,從而產生二倍體的雄性和雌性配子。

TDM屬於植物中保守的小家族蛋白。例如,擬南芥Arabidopsis基因組含有與TDM顯示出顯著序列相似性的5個其他基因(圖2)。這些類TDM基因的功能是未知的。蛋白序列的分析表明,因果突變在僅TDM蛋白亞家族中保守的小結構域中,所述TDM蛋白亞家族通常每個植物物種含有一個或兩個基因(圖1)。Pro17胺基酸以及相鄰的Thr16胺基酸是絕對保守的(圖1)。這就限定了T16上的最小共有磷酸化位點。為了對此進行驗證,通過用不可磷酸化的胺基酸(TDM-T16A)來置換可磷酸化的胺基酸,並通過刪除整個保守結構域(TDM-Δ14_19)來建立基因組TDM基因的兩個其它潛在的磷酸化丟失形式。當引入到野生型植物中時,TDM-T16A和TDM-Δ14_19都以顯性方式產生了二分體表型,重現了TDM-P17L的作用(表II;圖4)。此外,當引入到tdm-3突變體時,TDM-Δ14_19也顯示二分體表型(表II)。然而,當引入到野生型時,TDM酪氨酸14(TDM-Y14A)(結構域的保守性稍低的胺基酸)的突變不能產生二分體表型,但能夠補充tdm-3突變(表II)。總之,TDM-P17L、TDM-T16A和TDM-Δ14-19突變的表達同樣能夠顯性地提早退出減數分裂。由於TP是潛在的磷酸化位點,這個結果表明TDM可能受到磷酸化的調節以確保減數第一次分裂至減數第二次分裂的過渡。
