1.用含10%小牛和的RPMI-1640培養液將K562細胞配成1&time105/ml濃度。用同樣培養液將MC配成1&time106/ml濃度。
2.取96孔圓底，每孔加0.1ml K562細胞懸液;每孔加適量MC使效靶比為1：1～5：1，每種處理3個復孔;3個對照孔只加效應細胞;8個對照孔只加靶細胞;4個對照孔只加細胞培養液。每孔加20μl Alamar Blue;每孔總量為0.2ml。
3.在37℃ 5% CO2的飽和水汽二氧化碳培養箱中培養24小時。
4.直接在螢光檢測儀上檢測螢光強度，激發波長530nm，發射波長590nm。各孔螢光強度值應減去培養液對照螢光強度的平均值，各復孔螢光強度的平均值記為AF。按下式計算特異性殺傷百分比：
